The Heads of House are responsible for all the Hogwarts students that belong to the House they represent. Some Heads openly favour students from their House, such as Severus Snape; others are fairer, displaying favouritism on occasion, such as Minerva McGonagall (PS8).

J. K. Rowling has stated that all Heads of House were once Sorted to that House.

The prerogative of expelling students belongs to the Heads of House and the headmasters (CS5). When Hogwarts is under the intervention of the Ministry of Magic, the High Inquisitor also has that power (OP19).

Before the Battle of Hogwarts, when Harry asks McGonagall to create a distraction so that he can search for Horcruxes, she uses Patronuses to summon other Heads of House. Flitwick and Sprout arrive in time to assist McGonagall in her duel against Severus Snape, but the headmaster manages to flee. Each of them is in charge of calling their students to the Great Hall, and of arranging their transportation out of the school—via Room of Requirement—if they are not of age or willing to stay and fight for the castle (DH30).

Heads of House in canon

Gryffindor

Albus Dumbledore was the Head of Gryffindor when Tom Marvolo Riddle studied at Hogwarts (CS13). When he took on the position of headmaster, McGonagall became Deputy Headmistress as well as Head of Gryffindor.

After Dumbledore's death, McGonagall calls Rubeus Hagrid along with the other three Heads of House to her office (HBP29). He does not take the post, however, for Severus Snape becomes Headmaster the following term. McGonagall is Head of Gryffindor for the school year of 1997 (DH30).

Hufflepuff

Pomona Sprout, the Herbology teacher, is the Head of Hufflepuff (GF36, DH30).

Ravenclaw

Filius Flitwick, the Charms teacher, is the Head of Ravenclaw (DH30).

Slytherin

Horace Slughorn was the Head of Slytherin before Severus Snape became part of the staff. He returned to the post after Snape murdered Dumbledore (HBP27); he led the Slytherins to Albus' burial (HBP30). Slughorn remained in that position the following year, with Snape as Headmaster (DH30).
