A pure-blood is technically a witch or wizard whose four grandparents were also witches and wizards. Several old pure-blood families, however, consider someone to be so only if they come from a long lineage "untainted" with Muggle blood. This does not correspond with the reality of any families currently in the wizarding world; they merely erase Muggles and Squibs from their family trees [1]. Nevertheless, most wizards acknowledge the need to marry Muggles and Muggle-borns in order to keep the wizarding world extant.
In a way, pure-blooded families are split into two currents: some, such as the Blacks, who take their status seriously and consider all others to be inferiors, and those, like the Weasleys, who are open-minded and welcoming of wizards with Muggle heritage. Those in the first group consider wizards and witches belonging to the section to be blood traitors.
Ancient families have undeniable status in the wizarding world and, in their perspective, are undoubtedly superior to both blood traitors and "Mudbloods". Most often their consider their lineage's traditions to be a great source of pride, not hesitating before expelling from the family individuals who do not agree with this point of view; the Black family is full of such examples (see Black Blood Traitors).
Even though J. K. Rowling has stated that she inspired herself in the Nazi ideals to come up with how Death Eaters see Muggles and wizards of Muggle heritage, it's important to notice that not all self-conscious pure-blood wizards are evil. They may even, as Sirius points out in OP6, believe that Voldemort has some interesting ideas, but it's possible that some felt personal restrictions against the ethnical cleansing. We see no example of this behaviour in the books, however.
